Wayne County route numbers consist of 3 digits, and are posted on intersection blade signs, as shown above (older signs omitted the "RD" designation). Even numbers are N-S roads, while odd numbers are E-W, and the routes are numbered sequentially from west to east. Not all consecutive numbers are used, however, as gaps are left in the sequence for the possible addition of more routes.

In addition, the numbering scheme is divided into three geographic swaths: numbers beginning in 1 are located north of Ridge Road (the former US 104 alignment comprising CRs 103, 143 and 163), numbers beginning with 2 are between this and NY 31, and those beginning with 3 are south of NY 31. On any route that crosses from one zone to another, the first digit is changed while the latter two are retained (with the exception of CR 200, which extends a short distance north of Ridge Road). In most cases, county routes correspond to a single road name, so that several route numbers may be strung along one continuous highway. On the other hand, a single name and number will often follow a series of turns to form a continuous path among several intersecting road segments.

Wayne County highways mostly comprise primary through roads and connectors between them, but most of the roads in the county don't fit this description, and so town highways make up the bulk of the road mileage.

In the "Towns" column, Villages are also listed. A short stretch of CR 143 is the only instance of any Wayne County highway entering a village.
